ABSTRACT

Objective: To investigate the clinicopathological features and immunohistochemical phenotypes of gastric SMARCA4-deficient undifferentiated carcinoma, and to discuss the daily diagnostics of this entity and analyze its prognosis. Methods: The cases of gastric SMARCA4-deficient undifferentiated carcinoma diagnosed at the Department of Pathology, Peking University Cancer Hospital, China from January 2010 to August 2022 were collected. The histological sections were reviewed, the immunohistochemical results and clinicopathological features were analyzed, and relevant literature was reviewed. Results: Pure foci of undifferentiated carcinoma were seen in 7 cases, and 1 case was accompanied by a moderately differentiated tubular adenocarcinoma component. Undifferentiated carcinoma foci showed similar sheet-like or solid diffuse growth pattern, medium-sized tumor cells characterized by 1-2 nucleoli, and abundant cytoplasm and rhabdoid appearance. The average patient age was 65±8 years. Six patients were male and 2 were female. Immunohistochemical staining showed that undifferentiated carcinoma of all 8 tumors were negative for SMARCA4 (BRG1). Among 7 patients who underwent SMARCA2 (BRM) and SMARCB1 (INI1) staining, 4 cases showed loss of BRM expression, 2 cases showed weakly positive staining, and 1 case was diffusely positive, but all 7 cases were diffusely strong positive for INI1. The neuroendocrine marker, synaptophysin, was weakly positive in 5 cases, while CgA and CD56 were negative in 8 cases. Ki-67 index was more than 70%. Two cases were mismatch repair deficient and showed the loss of MLH1/PMS2 expression, while 1 case showed only MSH2 loss. PD-L1 staining showed that combined positive score (CPS)≥1 in 4 cases (CPS ranging from 1 to 55) and CPS<1 in the other 3 cases. Four patients had clinical stage Ⅳ disease. Two of them died within 3 months after diagnosis. Conclusions: Gastric SMARCA4-deficient undifferentiated carcinoma/rhabdoid carcinoma is a rare group of highly malignant tumors with a poor prognosis. Loss of the core subunit of SWI/SNF complex may be associated with the development of dedifferentiated histological pattern and aggressive tumor progression, which may be more frequently accompanied with mismatch repair deficiency.

ABSTRACT

Objective:To explore the chain mediating effect of core self-evaluation and social withdrawal on the relationship between peer relationship and depression symptoms of adolescents. Methods:From September 2020 to October 2020, a sample of 1 936 students from grade 4 to grade 9 of different schools completed a cross-section questionnaire survey including the inventory of parent and peer attachment, core self-evaluation scale, child social preference scale-R and depression self-rating scale for children.SPSS 21.0 and SPSS PROCESS macro program were used for data statistics. The statistical methods included analysis of variance, correlation analysis and intermediary effect test.Results:(1) Peer relationship (94.78±17.27) was positively correlated with core self-evaluation (34.14±7.52) ( r=0.50, P<0.01), and negatively correlated with depression (12.21±6.02) and social withdrawal (32.34±11.45) ( r=-0.55, -0.58, both P<0.01). Core self-evaluation was negatively associated with social withdrawal and depression symptoms ( r=-0.48, -0.67, both P<0.01), while social withdrawal and depression was positively correlated ( r=0.54, P<0.01). (2) Peer relationship had a significant direct effect on depression symptoms (the effect value=-0.205, P<0.01). Core self-evaluation and social withdrawal respectively separate mediated the effect of peer relationship on depression symptoms (the effect value=-0.231, -0.088, 95% CI=-0.261--0.202, -0.110--0.068), while the chain mediating effect of the two was significant, and the effect value was -0.025(95% CI=-0.033--0.019). Conclusion:Good peer relationship can lead to higher core self-evaluation and less social withdrawal behaviors in adolescents that serves as buffer from depression.

ABSTRACT

Nano-LC-MS/MS was used to analyze trypsin digested deer-horn gelatin( DCG) and deer-hide gelatin( DHG) samples. The glycopeptides in DCG and DHG were quantified by Label-free quantitative( LFQ) peptidomics,on the basis of which the glycopeptides with significant difference in DCG and DHG were determined. As a result,5 736 peptides were identified from DCG samples, including 213 galactosyl-hydroxylysine containing peptides( Gal-Hyl-peptides) and 102 glucosyl-galactosyl-hydroxylysine containing peptides( Glc-Gal-Hyl-peptides),while 6 836 peptides were identified from DHG samples, among which there were 250 Gal-Hyl-peptides and 98 Glc-Gal-Hyl-peptides. With over 3-fold peak area difference and highly significant intergroup difference( P < 0. 01) as the screening criteria,444 differential peptides were determined in DCG and DHG,including 16 Gal-Hyl-peptides and 5 Glc-Gal-Hyl-peptides. Then XIC peak shapes, standard deviation of peak area, and fold change were applied for further screening and 5 glycopeptides with significant differences in DCG and DHG were confirmed, which could serve as potential biomarkers for distinguishing DCG and DHG. The present study provided ideas and strategies for the in-depth investigation on the discrimination of DCG and DHG and is of good theoretical significance and application value for the further research on chemical constituents and quality control of gelatin derived Chinese medicinals.

ABSTRACT

OBJECTIVE: To analyze the distribution characteristics of surveillance results of key occupational diseases in Nanning City.METHODS: The occupational health monitoring data of key occupational diseases of Nanning City from 2015 to 2019 were collected using judgment sampling method and analyzed by descriptive epidemiology method. RESULTS: A total of 38 cases of key occupational diseases were reported in Nanning City during the past 5 years. The main diseases were occupational pneumoconiosis and occupational noise-induced deafness(ONID). The rate of occupational health examination was 40.06%, showing an increasing trend with the increase of years(P<0.01). The detection rate of occupational pneumoconiosis was higher than that of ONID(0.19% vs 0.01%, P<0.01). However, the abnormal detection rate of occupational health special examination in noise-exposed workers was higher than that in dust-exposed workers(10.98% vs 0.35%, P<0.01). The detection rate of pneumoconiosis-like changes in dust-exposed workers was the highest in private enterprises(P<0.01). The detection rate of binaural high frequency average hearing threshold ≥40 dB was highest in small and micro enterprises and private enterprises(all P<0.01). The detection rate of blood lead level >400 μg/L was 24.75%, and 98.80% of the abnormal workers were concentrated in medium-sized foreign-funded enterprises. The detection rates of abnormal leukocyte, neutrophil and platelet counts in benzene-exposed workers were 1.17%, 3.21% and 0.26% respectively. CONCLUSION: Among the key occupational disease risk factors in Nanning City, the number of workers exposed to dust and noise is relatively high, which results in serious consequences and harm. But the hazards of lead and benzene cannot be ignored. Emphasis should be placed on strengthening the supervision and management of key occupational diseases in small and medium-sized micro-enterprises and private enterprises.

ABSTRACT

BACKGROUND@#The catheter-tissue contact force (CF) is one of the significant determinants of lesion size and thus has a considerable impact on the effectiveness of ablation procedures. This study aimed to evaluate the impact of CF on the lesion size during right ventricular outflow tract (RVOT) ablation in a swine model.@*METHODS@#Twelve Guangxi Bama miniature male pigs weighing 40 to 50 kg were studied. After general anesthesia, a ThermoCool SmartTouch contact-sensing ablation catheter was introduced to the RVOT via the femoral vein under the guidance of the CARTO 3 system. The local ventricular voltage amplitude and impedance were measured using different CF levels. We randomly divided the animals into the following four groups according to the different CF levels: group A (3-9 g); group B (10-19 g); group C (20-29 g); and group D (30-39 g). Radiofrequency ablations were performed at three points in the free wall and septum of the RVOT in power control mode at 30 W for 30 s while maintaining the saline irrigation rate at 17 mL/min. At the end of the procedures, the maximum depth, surface diameter, and lesion volume were measured and recorded. A linear regression analysis was performed to determine the relationship between continuous variables.@*RESULTS@#A total of 72 ablation lesions were created in the RVOT of the 12 Bama pigs. The maximum depth, surface diameter, and volume of the lesions measured were well correlated with the CF (free wall: β = 0.105, β = 0.162, β = 3.355, respectively, P < 0.001; septum: β = 0.093, β = 0.150, β = 3.712, respectively, P < 0.001). The regional ventricular bipolar voltage amplitude, unipolar voltage amplitude, and impedance were weakly positively associated with the CF (β = 0.065, β = 0.125, and β = 1.054, respectively, P < 0.001). There was a significant difference in the incidence of steam pops among groups A, B, C, and D (free wall: F = 7.3, P = 0.032; septum: F = 10.5, P = 0.009); and steam pops occurred only when the CF exceeded 20 g. Trans-mural lesions were observed when the CF exceeded 10 g in the free wall, while the lesions in the septum were non-trans-mural even though the CF reached 30 g.@*CONCLUSIONS@#CF seems to be a leading predictive factor for the size of formed lesions in RVOT ablation. Maintaining the CF value between 3 and 10 g may be reasonable and effective for creating the necessary lesion size and reducing the risk of complications, such as steam pops and perforations.

ABSTRACT

Objective Melanoma is a malignant tumor with high clinical malignancy, rapid progression, poor prognosis and high mortality. It is of great clinical significance to explore the molecular mechanism of high metastasis of melanoma and find new tumor markers and therapeutic targets. The expression of mitochondrial fusion protein-2 (MFN2) in cutaneous melanoma tissues and peritumoral tissues was studied, and its correlation with clinical parameters was analyzed. The correlation between MFN2 and the biological behavior of melanoma was also discussed. Methods Immunohistochemistry was used to detect the expression of Med19 protein in cutaneous melanoma tissues and peritumoral tissues, and to analyze the correlation between MFN2 expression and clinical factors in patients. The lentiviral vector containing MFN2 coding sequence (Lenti-MFN2) infected with melanoma cell B16 was set up as the study group, and the lentivirus with green fluorescent protein gene (Lenti-GFP) as the control group. The mRNA and protein expressions of MFN2 and Ras-Raf1-ERK1/2 signaling pathways in transfected cells were detected by real-time quantitative PCR (qRT-PCR) and Western blot. Flow cytometry was used to detect changes in cell cycle and apoptosis. MTT assay and colony formation assay were used to detect changes in cell viability and proliferation capacity. Results The positive expression rate of MFN2 in peritumoral tissues was significantly higher than that in the skin melanoma tissues (83.9% vs 30.6%, P<0.05). MFN2 expression was associated with both lymph node metastasis and TNM staging (P<0.05). Compared with the control group, the proliferation activity and colony forming ability of B16 cells in the study group were significantly inhibited (P<0.05); the proportion of G0/G1 phase cells in B16 cells in the study group significantly increased [(46.3±10.3)% vs (67.9±12.6)%], and the apoptosis rate significantly increased [(12.5±1.6)% vs (57.4±12.6)%], with statistically significant differences (P<0.05). In comparison with the control group, the expression of Ras, Raf, ERK1/2 mRNA and protein in the study group significantly decreased (P<0.05). Conclusion MFN2 is down-regulated in melanoma tissues. Up-regulation of MFN2 expression can inhibit melanoma proliferation and promote cell apoptosis, which may be related to the inhibition of Ras-MAPK signaling pathway activity.

ABSTRACT

BACKGROUND@#Classification of the pulmonary neuroendocrine tumor (pNET) categories is a step-wise process identified by the presence of necrosis and number of mitoses per 2 mm. In neuroendocrine tumor pathology, Ki-67 was first described as a prognostic factor in the pancreas and incorporated into the grading system of digestive tract neuroendocrine neoplasms in the 2010 WHO classification. However, the significance of Ki-67 in pNETs was still a controversial issue. This study was to investigate the potentially diagnostic value of Ki-67 in pNETs.@*METHODS@#We retrieved 159 surgical specimens of pNETs, including 35 typical carcinoids (TCs), 2 atypical carcinoid (ACs), 28 large-cell neuroendocrine carcinomas (LCNECs), 94 small-cell lung cancers (SCLCs). Manual conventional method (MCM) and computer-assisted image analysis method (CIAM) were used to calculate the Ki-67 proliferative index. In CIAM, 6 equivalent fields (500 × 500 μm) at 10× magnification were manually annotated for digital image analysis.@*RESULTS@#The Ki-67 index among the 4 groups with ranges of 0.38% to 12.66% for TC, 4.34% to 29.48% for AC, 30.67% to 93.74% for LCNEC, and 40.71% to 96.87% for SCLC. The cutoff value of Ki-67 index to distinguish low grade with high grade was 30.07%. For the univariate survival analyses in pNETs, both the overall survival and progression-free survival correlated with Ki-67 index. In addition, the Ki-67 index performed by CIAM was proved to be of great positive correlation with MCM.@*CONCLUSIONS@#Ki-67 index counted by CIAM is a reliable method and can be a useful adjunct to classify the low- and high-grade NETs.

ABSTRACT

Objective To explore the source of human infection H9N2 avian influenza virus (AIV). Methods Environmental AIV nucleic acid monitoring was conducted for live poultry markets in Changsha city from 2014 to 2015, and data of human infection H9N2 subtype AIV cases worldwide were collected. Phylogenetic trees of hemagglutinin(HA), neuraminidase(NA)and non-structural protein(NS)genes from human infection H9N2 subtype AIV, the live poultry markets environmental H9N2 subtype AIV and partial avian H9N2 subtype AIV were constructed using the MEGA 6.06 software, respectively. Results In 2014-2015, H9 subtype AIV had the highest nucleic acid positive rate (44.76%) in the live poultry markets environment of Changsha city, and the pollution was serious. A total of 27 cases of human infection with H9N2 subtype AIV had been reported worldwide, and most of these patients recovered after treatments.Epidemiological survey showed that 59.26% (16/27) of cases had a clear history of exposure to poultry or live poultry markets. The phylogenetic trees of HA, NA and NS genes showed that the human infection H9N2 subtype AIV isolates isolated from Hunan and Guangdong were closely related to the H9N2 subtype AIV isolated from the live poultry markets environment in Hunan and Guangdong provinces from 2013 to 2016. The nucleotide similarity was as high as 97%-99%. Conclusion Live poultry market is one of the sources of human infection with H9N2 influenza virus.

Among 199 diarrheagenic E .coli strains ,the rates of resistance to ampicillin ,tetracycline ,trimethoprim-sulfamethoxazole ,azithromycin ,and ceftriaxone were 63 .8% ,55 .8% ,48 .2% ,34 .2% and 26 .6% , respectively ,while the rates of resistance to ciprofloxacin , amoxicillin-clavulanate and cefoxitin were 4 .5% ,1 .5% and 0 .5% ,respectively .Conclusions Diarrheagenic E .coli is the most common enteric bacteria detected in the stool samples from children with and without diarrhea in this study . The pathogenic role of infections with EPEC ,EAEC and ETEC in childhood diarrhea is not determined .EHEC and EIEC are rarely detected and further studies are needed to clarify the pathogenic association between infection with EHEC ,EIEC and childhood diarrhea .

ABSTRACT

Objective To compare the clinical efficacy and safety with statin and ezetimibe in the treat-ment of ACS after PCI. Methods 126 patients with acute coronary syndrome(ACS)who underwent percutaneous coronary intervention(PCI )were divided into three groups according to the lipid-lowering strategy. group A:mod-erate-intensity statin group(n = 47),group B:Ezetimibe combined with moderate-intensity statin(n = 38), group C:Fortified statin(n = 41). The levels of serum lipids, liver enzymes, creatinine, creatine kinase(CK), and the compliance ratio of lipids were compared between the three groups at 1 month and 6 months after PCI re-spectively. All patients were followed up for 12 months, then reviewing coronary angiography and comparing MACE incidents within 12 months. Results The levels of cholesterol in group A,group B and group C decreased signifi-cantly(P < 0.05)at one month after PCI,the percentage of decrease of TC in three groups was 18.95%,35.61%and 19.84% and 24.89%,and LDL-C was 39.56%,23.99%,respectively. The percentage of TC and LDL-C in group B were significantly lower than those in other two groups(all P < 0.05). One months after PCI,the compli-ance rate of LDL-C in group B(65.8%)was higher than that in A group(34.0%)and C group(31.7%),P <0.05. Six months after PCI,the compliance rate of LDL-C in group B( 91.3% )was still significantly higher than that in group A(79.4%)and group C(84.2%),but P > 0.05. The incidence of MACE in group B was significant-ly lower than that in group A and C(P < 0.05)at 12 months after PCI,while the incidence of side effects in group C was significantly higher than that in group B and group A(P < 0.05). The incidence of neovascular steno-sis in group B was lower than that in group A and C(all P < 0.05). There was no significant difference in the intra-ventricular restenosis among the three groups(all P > 0.05). Conclusions Ezetimibe-statin combination therapy reduced TC and LDL-C levels more significantly than statin alone(including moderate-intensity statin therapy and strengthen statin therapy),and the compliance rate of LDL-C at 1 month after PCI was significantly higher than that of statin alone. The incidence of MACE was lower in combination group than in statin group at 12 months after operation, and the side effects were less. Above all,the combination of statin and ezetimibe may be the secondary prevention for CHD in patients with acute coronary syndrome after PCI.

ABSTRACT

Objective To establish an animal model of Sparganum mansoni (plerocercoid larva of S. mansoni) infection in mice and observe the changes of blood routine examinations and serum anti-sparganum antibody levels after the infection. Methods The spargana tapeworms were collected from frogs, and 25 Kunming mice were orally infected with the Sparganum tapeworms (3 tapeworms/mouse). Two days before the infection and 2, 7, 14, 21, 28, 35, 42 days and 49 days after the infection, the peripheral blood samples of mice were collected for the blood routine examinations and the detections of anti-S. mansoni IgG antibody with ELISA. Forty-nine days after the infection, all the mice were sacrificed to find out the Sparganum tapeworms in the bodies of mice. Results The count of the total white blood cells was significantly elevated on the second day of the mice infected with Sparganum. The serum anti-Sparganum antibody was detected on the 14th day of the infection in some mice, and on the 21st day of the infection, the serum anti-Sparganum antibody was detected in all the mice. After the mice were sacrificed, the Sparganum tapeworms were found out in many tissues and organs, and especially in the subcutaneous tissues and muscle. Conclusion The establishment of animal model of Sparganum infection is successful in mice with the oral method, and white blood cells and serum specific IgG antibody detection can be used as auxiliary diagnosis methods of S. mansoni infection.
